Syntab Therapeutics is advancing a pipeline of innovative immuno-oncology drug candidates based on its proprietary ISEr platform. The platform enables the development of mono-, bi- and multispecific molecules designed to direct therapeutic activity toward defined molecular targets and modulate key immune mechanisms involved in cancer. It supports the rapid identification and development of binders against established targets as well as emerging targets with therapeutic potential.

Our development pipeline is led by two complementary candidates: ISEr-Y9, the pilot candidate that established the initial proof of concept, and ISEr-X9, our lead candidate advancing through mechanistic validation and translational development. Together, they demonstrate the progression of the ISEr platform from initial validation toward the development of a differentiated immunotherapeutic approach for solid tumors, such as Triple-negative breast cancer (TNBC).
TNBC is an aggressive breast cancer subtype defined by the absence of estrogen and progesterone receptors and HER2 overexpression or amplification. Accounting for approximately 15–20% of breast cancer cases, TNBC is associated with substantial risks of early recurrence and metastatic progression. Although treatment options have expanded to include immunotherapy and selected targeted approaches, significant unmet medical needs remain, particularly for patients with advanced disease or limited therapeutic options.
Syntab is developing a differentiated therapeutic approach targeting Integrin α3 (ITGA3), a cell-surface molecule of interest in tumor biology. By combining target-directed recognition with immune-modulating activity, the ISEr platform aims to engage tumor-associated biology while activating immune mechanisms that may contribute to tumor control.
ISEr-Y9 is Syntab Therapeutics’ pilot candidate and was developed to establish the initial proof of concept for the ISEr platform. Directed against Integrin α3, ISEr-Y9 has demonstrated promising antitumor activity in preclinical studies, including tumor-growth inhibition and prevention of metastatic progression in experimental models.
These findings provided an important foundation for the platform’s target-directed therapeutic approach and informed the subsequent development of ISEr-X9. ISEr-Y9 is also supported by Syntab’s intellectual property portfolio.
ISEr-X9 is Syntab Therapeutics’ lead immuno-oncology candidate, being developed for ITGA3-positive solid tumors, initially focusing on triple-negative breast cancer.
Derived from the proprietary ISEr platform, ISEr-X9 is a palmitoylated, bispecific molecule that combines tumor-directed targeting with the dual activation of formyl peptide receptors (FPR) and Toll-like receptors 2/1 (TLR2/1). This coordinated activity is designed to reprogram neutrophilic granulocytes toward an antitumor phenotype, harnessing innate immune mechanisms to promote tumor-cell elimination.
ISEr-X9 has generated promising in vitro and in vivo results supporting its continued development. Initial studies have demonstrated specific binding to 4T1 tumor cells and pronounced tumor necrosis in a syngeneic 4T1 tumor model. These findings provide a foundation for further mechanistic validation and translational development.
Syntab is advancing ISEr-X9 toward clinical translation, with the next development stage focused on strengthening the translational evidence needed to guide clinical evaluation.
Beyond TNBC, Syntab is investigating the potential application of ISEr-X9 to additional ITGA3-positive solid tumors, particularly lung and pancreatic cancer.
Our goal is to translate the promising preclinical findings for ISEr-X9 into a scientifically grounded clinical development program for patients with ITGA3-positive solid tumors.
